    Traditional vs. novel approaches to coastal risk management: A review and insights from Italy

    Coastal areas frequently face critical conditions due to the lack of adequate forms of land use planning, environmental management and inappropriate coastal risk management, sometimes leading to unexpected and undesired environmental effects. Risk management also involves cultural aspects, including perception. However, the acknowledgement of risk perception by stakeholders and local communities, as one of the social pillars of risk analysis, is often lacking.. Starting from an overview of the risk concept and the related approaches to be addressed, the paper investigates the evolution of coastal risk management with a focus on the Italian case study. Despite the design and adoption of national policies to deal with coastal risks, coastal management still shows in Italy a fragmented and poorly coordinated approach, together with a general lack of attention to stakeholder involvement. Recent efforts in the design of plans aiming at reducing risks derived from climate change and mitigating their impacts (National Strategy on Climate Change Adaptation; National Climate Change Adaptation Plan; National Recovery and Resilience Plan activities) should be effective in updating knowledge about climate change risks and in supporting national adaptation policies

    Plasma Concentration Rise after the Intramuscular Administration of High Dose Medetomidine (0.13 mg/kg) for Semen Collection in Cats

    High dose medetomidine 0.13 mg/kg can be used for semen collection in cats with variable results in terms of quantity and quality. Therefore, a variation in terms of distribution and elimination among patients has been hypothesised. The aim of the study was to characterise the pharmacokinetics of medetomidine (0.13 mg/kg) administered intramuscularly (IM) in healthy male cats. Eighteen male cats undergoing castration were included, and medetomidine (0.13 mg/kg) was administered IM. Venous blood samples were collected at 20, 30, 40, 50, 60, 75 and 90 minutes after medetomidine administration. Before orchiectomy, at T20, sperm collection was attempted. Plasma medetomidine concentrations were determined by liquid chromatography/mass spectrometry analysis. Semen collection was successful in 15/18 cats. The medetomidine plasma concentration following the IM administration of a bolus was best described using a non-compartment model. Time of maximum concentration was observed at 40 minutes (range 20-90); maximum concentration was 32.8 ng/mL (range 26.8-51.2). The median apparent clearance was 11.9 mL/kg/minute (range 0.7-43.8). In conclusion, medetomidine administered IM at 0.13 mg/kg reached its peak plasma concentration slowly and with variability among patients. In addition, it was characterised by low total body clearance probably due to the cardiovascular alterations associated with medetomidine administration

    Sequential or Concomitant Inhibition of Cyclin-Dependent Kinase 4/6 Before mTOR Pathway in Hormone-Positive HER2 Negative Breast Cancer: Biological Insights and Clinical Implications

    About 75% of all breast cancers are hormone receptor-positive (HR+). However, the efficacy of endocrine therapy is limited due to the high rate of either pre-existing or acquired resistance. In this work we reconstructed the pathways around estrogen receptor (ER), mTOR, and cyclin D in order to compare the effects of CDK4/6 and PI3K/AKT/mTOR inhibitors. A positive feedback loop links mTOR and ER that support each other. We subsequently considered whether a combined or sequential inhibition of CDK4/6 and PI3K/AKT/mTOR could ensure better results. Studies indicate that inhibition of CDK4/6 activates mTOR as an escape mechanism to ensure cell proliferation. In literature, the little evidence dealing with this topic suggests that pre-treatment with mTOR pathway inhibitors could prevent or delay the onset of CDK4/6 inhibitor resistance. Additional studies are needed in order to find biomarkers that can identify patients who will develop this resistance and in whom the sensitivity to CDK4/6 inhibitors can be restored

    3D Microfluidic model for evaluating immunotherapy efficacy by tracking dendritic cell behaviour toward tumor cells

    Immunotherapy efficacy relies on the crosstalk within the tumor microenvironment between cancer and dendritic cells (DCs) resulting in the induction of a potent and effective antitumor response. DCs have the specific role of recognizing cancer cells, taking up tumor antigens (Ags) and then migrating to lymph nodes for Ag (cross)-presentation to naïve T cells. Interferon-α-conditioned DCs (IFN-DCs) exhibit marked phagocytic activity and the special ability of inducing Ag-specific T-cell response. Here, we have developed a novel microfluidic platform recreating tightly interconnected cancer and immune systems with specific 3D environmental properties, for tracking human DC behaviour toward tumor cells. By combining our microfluidic platform with advanced microscopy and a revised cell tracking analysis algorithm, it was possible to evaluate the guided efficient motion of IFN-DCs toward drug-treated cancer cells and the succeeding phagocytosis events. Overall, this platform allowed the dissection of IFN-DC-cancer cell interactions within 3D tumor spaces, with the discovery of major underlying factors such as CXCR4 involvement and underscored its potential as an innovative tool to assess the efficacy of immunotherapeutic approaches

    Dendritic cells infiltrate the cardiac muscle fibers during myocardial infarction

    Myocardial infarction (MI) consists in myocardial cell death due to prolonged ischemia. Partial ischemia at the periphery at the necrotic area may lead to “hibernating” myocardium, which may eventually recover. Upon necrosis an inflammatory a process starts [1], leading to healing through formation of a fibrous scar. Dendritic cells (DC) are involved in the regulation of immune responses and in the organization of inflammatory cell infiltrates in vascular wall, even independent of immune reactions. Another cell type involved in acute reaction to tissue injury are mast cells. The behaviour of DC and mast cells in myocardial infarction is still to be studied. To address this issue myocardial samples were taken at autopsy from the left ventricle of subjects respectively affected by (1) coronarosclerosis, (2) acute MI, (3) previous MI, and (4) traumatic lesions assumed as controls. Cryosections were stained with haematoxilin heosin and by immunohistochemistry. Fiber alterations consisting in loss of acidophilia and disappearance of nuclei and intercalar disks were found only in acute MI, while a cell infiltrate was found both in acute and previous MI. Massive infiltration of DC was found only in acute MI, while mast cells were similar to controls. These preliminary results suggest that DC react early to myocardial injury and therefore may be candidate regulators of the inflammatory and scarring response in this tissue and markers of acute myocardial infarction
